I think that desmond is a big part of what might affect the others, first remember they other episode when they talked about the sub, the sub is back. And the end of last season, when Desmonds fiance got the call about the disturbance after desmond release the magnetic energy. I think that gave her the exact location of where they are and now the sub is trying to locate the Island.

I think “the sub’s back” meant that the team of Others who had gone over to the other island, the group that injured (now dead) Colleen was in, had returned, after Jin shot Colleen.
I also was assuming that the line was in reference to that group of people who went after the sailboat.
I do think that we will see an update on the attempt to find the island from the outside this season, but I don’t know whether this was it.
It’s more than obvious that Desmond’s power will ultimately be what gives the Survivors the upper hand against the Others.
I’m not sure how you got that impression. (We’re probably a long way off from anyone finding the island, even though Penny got a small clue.) A sub would explain how the Others boarded the sailboat to sneak up on Sun without making any noise and without being wet. There was no boat, they didn’t swim…they took a submarine. It might also explain how the Others get between the two islands.
No they take the tunnel for that.
The room Jack is in is an aquarium. He asked Juliette if they were underwater and she said yes… so there must be something underwater connecting the two islands.
